Summary:
It has been suggested that CYFRA21-1, a cytokeratin subunit 19 fragment, is potentially useful for diagnosis and monitoring of lung carcinoma. Serum levels of CYFRA21-1 are increased in a high proportion of patients with interstitial lung disease.
This assay employs the quantitative sandwich enzyme immunoassay technique. A monoclonal antibody specific for CYFRA21-1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CYFRA21-1 present is bound by the immobilized antibody: An Biotin-linked monoclonal antibody specific for CYFRA21-1 is added to the wells. Following a wash to remove any unbound Biotin-antibody enzyme reagent, Avidin-HRP is added to the wells, following a wash again; a substrate solution is added to the wells and color develops in proportion to the amount of CYFRA21-1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
